Installed Wine....but how exactly do I use it?
Do I have to have a Windows partition, or can it run Windows apps that have been downloaded onto a Linux partition, or what?

it depends on how you installed it.
if you used the source and use './tools/wineinstall' then it gives you the option to use your windows partition or to create your own fake window directories in linux.
you can install windows apps that are downloaded on your linux partition, but there are limits, check out http://www.winehq.com/ for more info on wine
