Can you tell us a little more about what your needs are and what you are trying to accomplish? There may be other ways to do it. Other than that, here is a link for Uboot manual.

Using Puppy Linux, 5.25, I have made several booting PenDrives and booting SD-cards.
For me, it has always been just a 'format', grub4dos install, and copy my existing Puppy 5.25 system files (all five of them)
onto the device. They always boot.
I have found that Puppy Linux will boot from almost any format (Fat32,NTFS,Ext2,Ext3).
RE: BIOS
There can be conflict between several USB devices during boot, all depending on the specific BIOS chip your system has installed.
On my Dell, it only boots from USB PenDrive if ONE bootable device is plugged in, and NO others plugged in.
On my HP, it boots if at least one is 'bootable', no conflict with any others plugged in.
Booting generally will not occur if the 'bootable' device is plugged into a 'hub' or a 'switch'.
