Why do you want to use a printer under wine? doesn't a native driver exist?
What distro are you using, and what printer are you trying to setup?Life is complex, it has a real part and an imaginary part.

So you want the system to start printing without asking you which printer to use? If that isn't the case, can you state more clearly what your intention is.

In the KDE control centre or Gnome control centre there is an option to set the default printer, have you tried setting it up there? You could also try setting it up in http://localhost:631
